Profiling of volatiles in the leaves of Lamiaceae species based on headspace solid phase microextraction and mass spectrometry

dc.description.abstractTo establish the volatile metabolomic profile of fresh leaves from Mentha × piperita L. and Mentha spicata L. aromatic plants, a headspace solid-phase microextraction (HS-SPME) technique combined with gas chromatography–mass spectrometry (GC–qMS) is reported. To assure the best extraction conditions, the influence of experimental parameters on the extraction efficiency, including the nature of fibre coatings, extraction temperature and extraction time, were investigated and optimized. The conditions that promoted the higher extraction efficiency, corresponding to the higher GC peak areas and number of identified metabolites were: DVB/CAR/PDMS coating fibre, 40 °C and 60 min as the extraction temperature and extraction time, respectively. Kovàts indices, mass spectra and/or standard compounds were used to identify up to 52 and 37 volatile organic metabolites (VOMs) in the headspace of M. × piperita L. and M. spicata L. fresh leaves, respectively, representing 91.26% and 88.52% of the total peak area of each plant. These VOMs belonging to different chemical families namely monoterpenes hydrocarbons, oxygenated monoterpenes, sesquiterpene hydrocarbons, oxygenated sesquiterpenes, pyrazines, higher alcohols, ethyl esters and carbonyl compounds. D-Carvone (54.77%) was the most abundant component isolated from the headspace of M. × piperita L. fresh leaves, followed by D-limonene (13.10%) and (+)-epi-bicyclosesquiphellandrene (5.14%), whereas M. spicata L. leaves contained 2,5-dimethyl-3-methoxypyrazine (54.58%), caryophyllene (5.50%) and D-germacrene (4.75%), as the main volatile metabolites. The results suggested that the HS-SPME/GC–qMS methodology could be used to establish the similarities and differences between both investigated Lamiaceae plants, and could probably be used for plant differentiation based on the volatile metabolomic profile.pt_PT
